HIP14 influences subcellular localization of SPRED1 and SPRED3 but not GPM6A in COS-7 cells. HIP14 substrates expressed alone or with HIP14-GFP in COS-7 cells were visualized by immunofluorescence microscopy. (A) FLAG-SPRED1 localized to the plasma membrane and colocalized with the Golgi marker GM130. Co-expression with HIP14-GFP promoted relocalization of FLAG-SPRED1 to vesicles. (B) Overexpression of FLAG-SPRED3 showed cytosolic and plasma membrane staining with no Golgi localization. Co-expression with HIP14-GFP promoted its colocalization in the Golgi. (C) FLAG-GPM6A localizes to the Golgi and to filopodia. No change was seen with HIP14-GFP co-expression; both proteins colocalized to the Golgi. Scale bar = 20 μm.
