Time course of cytokine-induced increase in proinsulin release. Cells were cultured in the presence (open circles) or absence (filled circles) of IL-1β (50 U/mL) plus IFN-γ (1,000 U/mL). Medium was sampled at 4, 24, and 48 hours and assayed for insulin and proinsulin. Data represent mean ± SEM of 3 independent experiments. Proinsulin release between 4 and 24 or 48 hours is significantly higher during cytokine exposure. Statistical significance of difference in controls was calculated by Mann-Whitney U test. *P < 0.05.
