CD4+ T cells recovered from NOD.scid.Il15−/− recipients do not inhibit proliferation of T cells or protect NOD mice from diabetes. (a) Splenocytes recovered from the reconstituted NOD.scid and NOD.scid.Il15−/− recipients were stained for CD4, CD25 and intracellular FOXP3. Expression of CD25 and FOXP3 in gated CD4+ cells was evaluated. (b) CD4+ T cells were purified from reconstituted NOD.scid, NOD.scid.Il15−/− and NOD.scid.gamma recipients and tested for their ability to suppress the anti-CD3-induced (CD4+ and 8.3 TCR Tg CD8+ T cells) and antigen-induced (8.3 T cells) proliferation by CFSE-dye dilution assay.
