BDC2.5 cells recovered from IL-15-deficient mice show reduced activation and IFN-γ production following stimulation with cognate antigen. (a) Splenocytes from NOD.scid, NOD.scid.Il15^−/− or NOD.scid.Il15ra^−/− recipients reconstituted with 0.5×10⁷ BDC2.5-NOD splenocytes were recovered 10 days later and labeled for the indicated markers. (b) The recovered splenocytes were activated with the cognate peptide (1 µg/ml) for 24 h. Upregulation of CD25 and CD69 and the expression of IFN-γ were determined on gated CD4⁺ cells.
