Elution of fractionations on cation exchange. A: Chromatography of the ultrafiltration fraction of 35% to 50% (NH4)2SO4 ammonium sulfate precipitation (22 mg of protein ) on CM-cellulose. The procedure was performed with a column (2.6 cm × 30 cm) at a flow rate of 65 to 75 mL per hour. About 5 mL fractions was pooled. The column was developed initially with 750mL of 0.011 M citric acid-NaOH (pH 4.5), 0.02% NaN3, and then a stepwise dilution over 400 mL from 0.05, 0.10, 0.15 to 0.25 mol/L NaCl was used to accomplish the development; B: Chromatography of the ultrafiltration fraction of 50% to 60% (NH4)2SO4 ammonium sulfate precipitation (5.6 mg of protein ) on CM-cellulose.
