Association between nickel contamination and in vitro measures of multi-walled carbon nanotubes (MWCNT) bioactivity in alveolar macrophages (AM). Freshly isolated C57Bl/6 AM (100 μl at 106 cells/ml) were incubated with 100 μg/ml of the nine MWCNT for 24 h. Measurement of IL-1β and IL-18 required costimulation with 20 ng/ml LPS to generate the pro-forms of the cytokines for the NLRP3 inflammasome-activated caspase-1 to process.
