NoV RNA replication is unaffected by the presence of a C-terminal HA epitope tag. (A and B) Total cellular RNAs were isolated from BSR-T7/5 cells transfected with either pT7-N1 (WT) or tagged pT7-N1-HA, separated on denaturing gels, and subjected to Northern blot hybridization analysis using probes specific for the positive (A) or negative (B) strands of RNA1, which also detect subgenomic RNA3. (C and D) Prior to transfer, gels were stained with ethidium bromide to allow visualization of 18S and 28S rRNAs for use as loading controls. (E to H) Quantitation of positive (+) and negative (-) strands of NoV RNA1 (N1) and NoV RNA3 (N3) relative to WT levels are shown: N1(+) (E); N1(-) (F); N3(+) (G); N3 (-) (H).
