Colocalization of mutated pUL34 with pUL31 after transient expression. In order to analyze the interaction between pUL31 and mutated pUL34, RK13 cells were cotransfected with the corresponding expression plasmids. Colocalization was studied using the monospecific murine anti-pUL34 (red) and rabbit anti-pUL31 (green) sera. Images were recorded by confocal laser scanning microscopy (Leica SP5). Speckles seen after coexpression of native pUL34 and pUL31 reflect the interaction of both proteins and the formation of primary envelopes (14), while diffuse nucleoplasmic pUL31 staining is due to failure of pUL31 recruitment. α, anti.
