Total cell lysates were isolated from 786-O or RCC4 (both naturally deficient of pVHL expression) cells stably expressing control vector or HA-tagged pVHL. Protein levels were analyzed by immunoblotting. B, accumulation of USP33 upon p97 knockdown in 786-O cells. 786-O cells expressing control vector or HA-pVHL were transduced with lentivirus encoding a doxycycline (dox)-inducible shRNA against p97 (p97-V1). Cells were treated with or without 1 μg/ml doxycycline for the indicated time, and total cell lysates were isolated and analyzed by immunoblotting.
