Adenoviral vectors encoding G5G8 (AdG5G8) reduces eukaryotic initiation factor 2α (eiF2a) signaling in db/db mice. Obese (db/db) mice were administered control and AdG5G8 vectors. A, Total and phosphorylated (P-) eiF2a, Akt, c-Jun N-terminal kinase (JNK), and pancreatic eiF2α kinase (PERK) were assessed in hepatic lysates by SDS-PAGE and immunoblotting. The activating transcription factor (ATF) 6 target genes glucose-regulated protein (GRP) 78 and GRP94 were also assessed. β-actin was used as a loading control. B, The ratios of phosphorylated to total eiF2a, Akt, and JNK were determined by densitometry. C, Reverse-transcriptase polymerase chain reaction was used to measure downstream signaling for eiF2a (ATF4, tribbles 3 [Trb3]), ATF6 (GRP78, X box–binding protein 1 [XBP-1]), and inositol-requiring enzyme 1 (spliced sXbp1).
