Adhesion on composite matrix. Wells were coated with equal concentrations of fibronectin and TFPI-1, as indicated on the x axis. (a) Effect of 10 nM VIIa on the adhesion to the composite matrix. The adhesion buffer contained 5 U/mL heparin to block proteoglycan-dependent cell adhesion to TFPI-1. (b) Effect of function-blocking anti-β1 integrin antibody on the binding to composite matrix. The antibody at 50 μg/mL completely blocked adhesion in the absence of VIIa (anti-β1, no VIIa). Adhesion to the composite matrix in the presence (+ anti-β1) or absence of the blocking antibody was analyzed at various concentrations of immobilized matrix and in the presence of 10 nM VIIa and 5 U/mL heparin.
