PC7 and furin affect HIF-1α protein level by a different mechanism than CoCl2. Cells were treated with cycloheximide (CHX) for 15, 30 and 60 minutes after HIF-1α induction under 3 different conditions: A. CoCl2 treatment overnight, B. PC7 SiRNA, and C. furin SiRNA transfection for 2 days respectively. The cell lysates were collected and analyzed by Western blotting for HIF-1α and GAPDH. D.
