MDA-231 (231) or MCF10A (10A) exosomes were fed to bone marrow-derived macrophages generated from indicated mouse strains. (a) IL-6 and (b) TNFα expression levels were determined by quantitative RT-PCR. *p < 0.001. (c) Endosomally localized TLRs in RawkB cells were inhibited by a 2-h pretreatment with indicated concentrations of Chloroquine. Stimulation was subsequently carried out by feeding cells with exosomes or TLR ligands for 4 h followed by assessment of luciferase activity. *p < 0.001 compared to the PBS (control) group. (d) RawkB cells were treated with MDA-231 exosomes or PBS in the presence of a TLR2 neutralizing antibody or a control antibody at a final concentration of 30 ng/ml.
