Mid-log phase cultures of wild type and ccr4Δ cells were kept at 30°C (A.) or shifted to 37°C (B.) in the presence of the transcriptional inhibitor 1,10-phenanthroline. Aliquots of cells were harvested at 15-minute intervals for RNA extraction and subsequent northern blotting for the detection of RPL2 and RPL27e transcripts. Bands corresponding to transcripts were normalized to rRNA and relative expression was plotted on a log2 graph to determine half-life. Gels and blots shown for RPL2 and RPL27e expression are representative of 3 biological replicates.
