Melan-INK4a cells were treated with siRNA oligos as indicated. A) PNS of cells transfected with the siRNA indicated were immunoblotted with antibodies indicated. B) Solubilised cell lysates were incubated with immobilised GST or GST-Slp1. Co-precipitation was assessed by immunoblotting for Rab27a and GST. The immunoblot is representative of 5 independent experiments. Depletion was quantified by measuring band intensities using ImageJ, normalised to GST-Slp1 and the protein quantified relative to NT siRNA control.
