Effects of Ang II and isoproterenol (ISO) on endogenous PDE3A expression in cardiomyocytes. Cardiomyocytes were stimulated with vehicle or Ang II (200 nmol/L) or isoproterenol (10 μmol/L) for indicated time. PDE3A and β-actin protein levels were measured by Western blotting (A, C), and PDE3A mRNA levels were determined by relative quantitative reverse transcription–polymerase chain reaction (B). D, Cardiomyocytes were treated with or without Ang II (200 nmol/L) or isoproterenol (10 μmol/L) for 72 hours in the presence or absence of losartan (10 μmol/L) or metoprolol (1 μmol/L). PDE3A and β-actin protein levels were measured by Western blotting.
