NHE5 regulates intracellular pH in AP-1 cells in response to metabolic stress-induced acidosis. A, untransfected AP-1 cells were treated with diluent or AA (1 μg/ml) over a 60-min period to inhibit oxidative phosphorylation and the effects on intracellular pH were measured using the ratiometric pH-sensitive fluorescent dye BCECF. B, AP-1/NHE5_HA3 cells were treated with diluent, AA (1 mg/ml), AA plus CC (20 μm), or AA plus HOE694 (1000 μm) over a 60-min period and the effects on intracellular pH were measured. The data are presented as the mean ± S.E. of at least 3 independent experiments.
