KLF4 is required for the action of BAY 60-6583. SVCs were isolated from the subcutaneous adipose tissue of WT mice. Cells received scrambled siRNA (Scrambled) or siRNA targeting KLF4 (KLF4 siRNA) and were treated with DMSO (Vehicle) or 1 μm BAY 60-6583 (BAY) on day 0 and day 3 of induction and induced to differentiate as described under “Experimental Procedures.” A–C, mRNA expression of KLF4 (A) at 0 h and PPARγ (B) and aP2 (C) after 3 days of differentiation was determined using the ΔΔCT method and normalized to 18 S rRNA values (n = 4). Data are mean ± S.E. *, p < 0.05; ***, p < 0.001 compared with scrambled (A) or vehicle treatment given the same siRNA (B and C).
