Felodipine inhibits Aurora A in a dose-dependent manner. (A) Aurora A (40 ng) was incubated with recombinant histone H3 and 2.5 μM [γ-³²P] ATP in the presence of an increasing concentration of felodipine (10, 20, 40, 60, 80, and 100 µM). In a similar experiment (B) histone H3 was incubated with Aurora B (40 ng) and [γ-³²P] ATP (lane 1), with DMSO, and with increasing concentration of felodipine (10, 20, 40, 60, 80, 100, 150, and 200 µM) and subjected to autoradiography. (C) Band intensity was quantified using a Fuji film PhosphorImager analyzer and plotted as a bar chart representing the extent of phosphorylation.
