Effect of PPARγ suppression on osteogenic differentiation of hADSCs. Human ADSCs were transfected with 50 nmol/l PPARγ2 siRNA for 48 hrs, and cultured in osteogenic induction medium for another 0, 2, 6 or 12 days (corresponding to 2, 4, 8 or 14 days after siRNA transfection) to determine the mRNA level of (A) BMP2, (B) Runx2, (C) ALP and (D) OC. (E) Cells were induced to differentiate osteogenically as described in Materials and methods.
