Tamoxifen toxicity causes lysosomal destabilization. (A) LAMP-1 (red) was localized to lysosomes in ARPE-19 cells by immunofluorescence in control cells (top left), and a progressive loss of LAMP-1 staining was observed with 30, 90 and 120 minutes of tamoxifen treatment (top right and bottom). Scale bar: 25 μm. (B) Quantification of LAMP-1 staining showed a statistically significant decrease in LAMP-1 fluorescence after 90 minutes, with no significant difference in cell counts when compared with the control. After 120 minutes, there was a statistically significant decrease in both LAMP-1 fluorescence and cell count as determined by DAPI+ nuclei. Error bars: ±SEM.
