p75 is a constitutive dimer in the membrane, stabilized by the Cys257 at the TM domain, where it can bind to NgR complex and activate RhoA signaling and axonal growth collapse. When p45 binds to p75 through both the TM domain (Cys257–Cys58) and DD interactions, p75 downstream signaling is inhibited.
