Western blot analysis of Grx1 and Grx2 content in the SSM from young adult and elderly Fischer 344 rat hearts. Heart SSM isolated from young adult and elderly F344 rats were treated with t-Bid to release the Grx1 localized in the mitochondrial intermembrane space. Subsequent mitochondrial fractionations resulted in “Supernatant,” “Mitochondrial Extract,” and “Residues” fractions, as described in detail previously [12]. Each fraction was analyzed for Grx1, Grx2, Ak-2 and cytochrome c by Western blot. The content of Grx1 and Grx2 was quantified by comparing the density of the bands to a standard curve generated on each gel using the corresponding purified human isozymes.
