RAB22A knockdown impairs breast cancer cell invasion. (A and B) A total of 2 × 105 cells of the MDA-MB-231 (A) and MDA-MB-435 (B) SC, sh22A-1, and sh22A-2 subclones were seeded on top of Matrigel-coated chamber inserts and incubated in serum-free DMEM under either 20% (N) or 1% (H) O2 for 24 h. The number of cells that invaded through the Matrigel to the underside of the filter was determined by staining with crystal violet and counting under bright field microscopy. (C and D) The number of invaded cells per field was determined from 10 fields per filter.
