FLAP plays a role in the cellular formation of LTC₄. (A) Differentiated control and knockdown MM6 cells (∼2 × 10⁶ cells in 1 mL of PBS) were incubated with 5 µM LTA₄ for 5 min at 25 °C. The formation of LTC₄ was determined by HPLC. Data are mean ± SD of two independent experiments; n = 6. ***P < 0.001. (B) Cell lysates of control and knockdown cells were subjected to Western blot analysis with anti-LTC₄ synthase antibody. The samples are supernatants of total lysates, and each sample contains 50 μg of protein. As a loading control, membranes were reprobed with β-actin to analyze the expression levels of LTC₄ synthase.
