Access to the perpendicular singlet excited state conformation is required for the trans to cis photoisomerization of stilbenes. When stilbenes are bound to or tethered to TTR (red curve), the perpendicular singlet excited state and the cis singlet excited state are strongly destabilized by the constraints imposed by the TTR binding site.
