Specific formation of ssDNA and RNA:DNA hybrids are observed in preparations of C. parapsilosis mtDNA. A, examples of ssDNA formation in both coding regions show that overall (almost always) the upper strand is detected as ssDNA. B, RNase-sensitive arcs are observed near the putative transcription start site around approximately nt 12,200. RNase 1 significantly reduces the structures, but RNase H removes them indicating that they are mainly RNA:DNA hybrids with RNA overhangs. Therefore we call them pseudo-RITOLS because they are most likely directly related to transcription, whereas an obvious connection to replication cannot be made.
