Upon the genotoxic stress, RAIDD favors the open conformation and binds to PIDD first via DD:DD interaction to allow PIDDosome formation. Interaction of PIDD to RAIDD stabilizes the open conformation of RAIDD, enabling caspase-2 to be recruited to the complex via CARD:CARD interaction.
