SecA variants were titrated onto a fixed concentration of MANT-labeled nucleotide at 25 °C in KEMT buffer (pH 7.6) while MANT fluorescence anisotropy was monitored at 352 nm. (A) WT (■) and E208Q (◆) BsSecA were titrated onto 900 nM Mg-MANT-ATP. (B) WT (□) and E210Q (◇) EcSecA were titrated onto 30 nM Mg-MANT-ATP.
