Echinocandin inhibition profiles of enriched GS complexes from the susceptible and resistant C. kefyr isolates. The in vitro inhibition of product-entrapped 1,3-β-d-glucan synthase (GS) complexes isolated from three C. kefyr strains was performed to determine the 50% inhibitory concentration (IC₅₀). Incorporation of [³H]glucose into the polymerized product was measured in GS isolated from one sensitive strain (C117) and two RES strains (C113 [R647G] and C116 [F641Δ]). The GS complexes prepared from isolates C116 and C133 were insensitive to echinocandins at up to 10,000 ng/ml. Error bars represent standard errors of the means (SEM).
