Radicicol-induced upregulation of P. falciparum topoisomerase VIB. Synchronized schizont-specific cultures were divided into two groups and grown for 40 h in the absence or presence of radicicol (1.5 μM). (A) Semiquantitative RT-PCR analysis of PfTopoVIB, PfHsp90, and PfARP genes in the absence (−) and presence (+) of radicicol. ARP acted as a loading control. PCR without reverse transcription (−RT) shows no amplification of PfTopoVIB. Genomic DNA (gDNA) sample acted as a positive control. (B) Real-time RT-PCR reveals quantitative abundance of the PfTopoVIB and PfHsp90 transcripts in radicicol-treated and -untreated cells. (C) Western blot analysis on parasite lysates isolated from radicicol-untreated (−) and -treated (+) cultures using anti-PfTopoVIB antibody. PfHsp70 acts as a loading control.
