NP is sumoylated in infected cells. 293T cells were cotransfected with FLAG-tagged SUMO1 or SUMO1AA and Ubc9 together with SENP1 or SENP1mut as indicated. At 24 h posttransfection, the cells were infected with the WSN-WT or WSN-NPK4,7R virus at an MOI of 1. The cells were lysed 16 h later, and SUMO1 was immunoprecipitated (IP) with anti-FLAG–agarose. The precipitated proteins were further analyzed by SDS-PAGE and immunoblotting (IB) with anti-NP monoclonal antibody. Immunoblot assays with anti-FLAG, anti-NP, or anti-actin polyclonal antibodies and whole-cell lysates are shown as SUMO1 expression, infectivity, and loading controls (Input), respectively. The values to the left of the blots are molecular sizes in kilodaltons.
