Proximity ligation detects binding of nearby epitopes. Protein is incubated with antibodies conjugated to oligonucleotides sequences. These antibodies bind to the protein(s), bringing the oligonucleotides sequences in proximity with each other. A connector oligonucleotide then facilitates ligation and replication through methods such as RT-PCR. A) Protein homodimerization results in nearby epitopes, causing oligonucleotide amplification. B) Protein heterodimerization results in nearby epitopes, causing oligonucleotides amplification. C) Detection of multiple epitopes on the same protein allows for increased sensitivity. Nearby binding to multiple epitopes causes oligonucleotide amplification.
