Detection of enzyme activity via substrate modification. A) Kinase phosphorylation of a substrate can be detected with phosphoantibodies. B) Protease cleavage of a substrate can be detected by the release of a fluorophore from a quenching molecule. C) In the telomere repeat amplification protocol (TRAP), telomerase extends oligonucleotides sequences by addition of the telomere sequence TTAGGG. This extension can then be detected by PCR amplification and polyacrylamide gel electrophoresis (PAGE).
