Principle of in situ hybridization. (1) Schematic organism with unstained adhesive organs. (2) The DIG-labelled RNA probe binds to the target mRNA and (3) is detected by an antibody conjugated with a phosphatase. (4) The former colorless substrate becomes dephosphorylated and turns blue. (5) The staining reveals the cells with target gene expression, in this case the adhesive organs.
