DQ2 does not show DM dependent conformational change during biosynthesis. 2.2.93, 9.5.3, and their DM transfectants (2.2.93-DM and 9.5.3-DM, respectively) were pulsed for 15 min with 35S-Met/Cys and then chased for 2, 2.5, 3, 3.5, 4, 4.5 hrs (lanes labeled as 1, 2, 3, 4, 5, 6, respectively), as described in Materials and Methods. Cell lysates (5 × 106 cell equivalents/lane) were immunoprecipitated with anti-DQ mAb, SPV-L3, and then analyzed by SDS-PAGE.
