Effect of capric acid on RANKL-induced osteoclast differentiation. BMMs were cultured with M-CSF (10 ng/ml) and RANKL (20 ng/ml) for 4 days at the indicated concentration of capric acid. (A) The cells were fixed and subjected to TRAP staining. (B) The number of TRAP-positive multinucleated cells (MNCs) generated in (A) was counted. (C) TRAP activity in the cell culture supernatant generated in (A) was measured by ELISA. The data presented in (B) and (C) are expressed as the means ± SD. *P < 0.05, **P < 0.001 versus vehicle-treated control (Con). (D) BMMs were cultured with M-CSF (30 ng/ml) for 3 days together with the indicated dose of capric acid. Cell viability was measured by an MTS assay, as described in “Materials and Methods”.
