Effect of capric acid on survival of mature osteoclast. Mature osteoclasts were treated with capric acid (500 μM) or vehicle for 24 h. (A) The cells were fixed and stained for TRAP. (B) Osteoclast survival was measured as the percentage of TRAP-positive MNCs. (C) The rate of osteoclast apoptosis was measured by ELISA. The data presented in (B) and (C) are expressed as the means ± SD. **P < 0.001. (D) The Bim content was determined by immunoblotting. β-Actin served as a loading control. (E) Cells were treated with capric acid (500 μM) or vehicle and then exposed to M-CSF (50 ng/ml) for 5 min. Total cell lysates were immunoblotted with an anti-phospho-ERK antibody. Total ERK served as a loading control.
