Neonatal male mice were injected intraperitoneally with either controlled saline or purified recombinant Lgr4-ED (10 µg/animal) on postnatal day 1, 8 and 15. Mice were then sacrificed on day 22 and (A) the testicular weights were measured. (B) The expression levels of Esr1, Esr2, Aqp1 in the harvested testes were compared by real-time PCR. Data were normalized against β-actin levels and represent the mean ± SEM (n = 6). Asterisks indicate P<0.05. (C and D) In addition, the testes were also subjected to immunohistochemical staining to detect the Esr1 protein and visualized using an alkaline phosphatase staining system.
