ABT-737 sensitizes primary B cells to differentiation-induced apoptosis. Primary murine B cells were treated with IL-4 and LPS for 96 h. ABT-737 was added during the final 24 h prior to collection. A and B, live cells determined by negative 7-aminoactinomycin D (7AAD) staining were gated into small B cells or large differentiating cells (representative experiment in A) and are quantified in B. C and D, total cells were gated into CD138-positive and CD138-negative fractions (C is representative of gating), and 72-h cell death was measured as the proportion of 7-aminoactinomycin D-positive (7AAD-pos.) cells (representative experiment in D) and is quantified in E.
