Splenic cells of control (untreated) and HEL- and protein L-treated transgenic mice were depleted of erythrocytes by using Tris-buffered ammonium chloride and stained with cell surface markers. (A) Total number of splenic cells counted by using a Malassez cell; (B) HuIgM+ B220+ cells as detected by PE-labeled anti-HuIgM and CyChrome-labeled anti-mouse B220 MAbs; (C) B220+ protein L+ cells as detected by FITC-labeled anti-mouse B220 MAb and biotinylated protein L, followed by treatment with CyChrome-labeled streptavidin.
