Figure 2.
sLZIP interacts with PPARγ2. (a) GST-sLZIP, Myc-PPARγ2, GST-PPARγ2, and Flag-sLZIP were co-transfected into 293T cells. Cell lysates were collected and GST pull-down assays were performed using glutathione 4B beads. (b) PPARγ2 was in vitro translated using a TNT translation reaction, and His-sLZIP proteins were expressed in BL21 cells. Purified His-sLZIP proteins were subjected to a His pull-down assay. (c) GFP-PPARγ2 and Flag-sLZIP constructs were expressed into C3H10T1/2 cells and analyzed by fluorescence microscopy using Texas Red-conjugated anti-Flag antibody. The nucleus was stained with DAPI. (d) 293T cells were transfected with GST-sLZIP (1-354), GST-sLZIP (1-228), GST-sLZIP (229–354), GST-sLZIP (297–354), and Flag-PPARγ2. (e) 293T cells were transfected with Flag-PPARγ2 (1-505), Flag-PPARγ2 (1-310), Flag-PPARγ2 (139-505), Flag-PPARγ1, and GST-sLZIP. GST pull-down assays were performed using glutathione 4B beads. Protein complex was analyzed on SDS-PAGE, followed by western blotting using anti-GST and anti-Flag antibodies
