B16-F10 cells were pre-treated with different concentrations of wogonin for 24 h. (A) Effects of wogonin on the protein levels of PI3K and PDK1 were examined. (B-C) The related proteins expression of NF-κB pathways (p-IKKα, IKKα, p-IκBα and IκBα) was tested. (D) Cytosolic fractions and nuclear extracts were prepared. Western blotting analyses was performed to investigate nuclear translocation of NF-κB p65. All protein was determined with specific antibodies. β-actin antibody was used to check the proper protein loading. Western blotting was done at least three times. *p<0.05 compared with control; **p<0.01 compared with control.
