Effects of BMP4 on activation of Smad1/5/8. (A) Cells were fixed and stained with anti-p-Smad1/5/8 and the nucleus was staining with 4',6-diamidino-2-phenylindole (DAPI). The phosphorylation of Smad1/5/8 was activated by BMP4 treatment in rat PASMCs, but the effect was eliminated by PI3K/AKT inhibitors, LY294002 and wortmannin. Scale bar = 10 µm; (B,C) Incubation of PASMCs with BMP4 led to phosphorylation of Smad1/5/8. The phosphorylation of Smad1/5/8 activated by BMP4 was partly inhibited by LY294002 and wortmannin (PI3K/AKT inhibitors). All values are denoted as means ± SEM from three or more independent batches of cells. “LY” means LY294002; “W” means wortmannin. * p < 0.05.
