Schematic view of Plasmodium falciparum GPI‐induced TLR2 signalling. TLR2 ECD recognizes GPIs either in combination with TLR1 or TLR6, then their TIR domains dimerize on the cytosolic face and undergo structural changes to recruit adaptor molecules: MyD88, MAL, IL‐1R‐associated kinases (IRAKs), (TGF‐β)‐activated kinase (TAK1), TAK1‐binding protein 1 (TAB1) and TAB2, and then tumour‐necrosis factor‐receptor‐associated factor 6 (TRAF‐6) forms a complex with ligases Ubc13 and Uev1A, which leads to the activation of the mitogen‐activated protein kinase and nuclear factor‐κB pathways to induce the production of proinflammatory cytokines.
