Deletion of Taf1p-region 4 dramatically reduces steady-state levels of holo-TFIID. WCEs prepared from the indicated yeast strains (Δ1Taf1p to Δ17Taf1p with HA₃ tag, WT with no HA₃ tag [−], and WT with HA₃ tag [WT] [Table 1]) were used for IP with monoclonal anti-HA antibody (MAb 12CA5). One percent of precleared lysate (Input) and 15% of each immunoprecipitate (Pellet) were analyzed by SDS-PAGE and immunoblotting with the antibodies recognizing the TFIID subunits indicated on the left.
