Knob-functionalized ELPBCs exhibited enhanced cellular uptake in CAR-expressing hepatocytes after 30 min incubation. Rhodamine-labeled block copolymers (Rh-S48I48) demonstrated significant localization at the cellular membrane, while rhodamine-labeled knob-functionalized block copolymers (Rh-knob-S48I48) demonstrated enhanced intracellular localization. Further evidence of enhanced cellular uptake was apparent in the colocalization of knob-functionalized ELPBC and a lysosomal marker. Green-LysoTracker; Red-ELPBC; Scale bars 10 µm.
