“Weak helper interaction” strategy for the generation of efficient FRET probes. (A) The FRET-based detection of a protein-protein interaction (PPI) between one protein of interest (POI1) and a second POI (POI2) is improved through the use of helper domains that bring the FP domains in closer proximity. Induction of a PPI leads to efficient FRET from mCitrine (cit) to mCherry (mCh). The helper interaction only occurs at high effective concentration resulting from the PPI of the POIs. (B) The helper interactions also improved the dynamic range of intracellular caspase sensors that use mTurquoise2 (mTu) and Citrine (cit) [98].
