Interaction of NdhO with NdhI and NdhK and between NdhI and NdhK. Yeast two-hybrid experiments to test the interaction of NdhO with various Ndh subunits (A) and between NdhI and NdhK (B). The ndhO and ndhI genes were constructed into bait vector, while each of the ndh genes encoding NDH-1M subunits except ndhO and a GST gene were constructed into a prey vector. Subsequently, they were transformed into the yeast strain EGY48. Transformed yeast was dropped onto X-gal medium. Blue precipitate represents accumulated β-galactosidase activity resulting from the activation of the lacZ reporter gene by protein-protein interaction. The induction plate was grown at 30 °C for 23 h and then photographed. The interaction of NdhI-GST was assayed as a negative control. At least six independent experiments were performed, and the result of one representative is shown.
