Sheep-adapted BSE-like but not scrapie-like PrPres is capable of converting human PrPC to PrPres. Human brain substrate of the PRNP codon 129 MM genotype was seeded at a 1:10 dilution with the initial (r1) and final round (r8) sPMCA products from an experiment in which sheep BSE isolates from VRQ/VRQ, ARQ/ARQ, and ARR/ARR sheep were seeded in ovine VRQ/VRQ (A), ARQ/ARQ (B), and ARR/ARR (C) brain substrates. Each sample was either not subjected to PMCA (−) or subjected to a single round PMCA for 48 h (+). Samples were analyzed after PK treatment using anti-PrP mAb 3F4. The molecular mass (in kDa) is marked to the left of each blot. Dotted lines indicate montaged individual Western blot results.
